Plant genetic improvement can be done through polyploidization. Polyploid taro Bentul (Colocasia esculenta L. Schott) has been produced from in vitro treatment of oryzalin 7,5-75 µM. The research was aimed to evaluate in vitro growth of polyploidy taro Bentul and stomata density of polyploid plants grown in the field. The level of ploidy was confirmed by flow cytometer. Stomata observation was done by light microscope. The results showed that in vitro growth of diploid and polyploid shoot cultures varied. Diploid taro had better growth than polyploid shoots. Hexaploid taro has slowed growth, more leaves (7,27 leaves per shoot), but had little size. Hexaploid taro had more roots than diploid and tetraploid taro (5,75 roots per shoot). The length, width, and density of stomata also vary between diploid and polyploidy. Density range of diploid taro stomata was higher compared to polyploid plants (more than 200 stomata per mm2).
